Exploitation of virus-like particles for vaccinology and the development of safe efficacious Bluetongue virus vaccine

Summary of the impact

Recent outbreaks across Europe of Bluetongue, a viral disease particularly affecting sheep, have driven research at LSHTM by Professor Polly Roy and her team, resulting in the Bluetongue virus (BTV) becoming one of the best understood viruses at the structural and molecular levels. The research has ultimately enabled the creation of several promising new vaccines. In addition the Roy group has contributed towards exploiting virus-like particles (VLPs) as a method to produce safe vaccines against human and animal viral pathogen. The most advanced example is a BTV vaccine for livestock, which is manufactured by Boehringer Ingelheim (BI).

Avian Metapneumovirus Research Leading to Improved Disease Control in Global Poultry

Summary of the impact

Since its discovery in the 1980s, avian metapneumovirus (AMPV) has spread in poultry populations worldwide with major adverse health and food security implications for commercial chickens and turkeys. Research at the University of Liverpool (UoL) led to the registration of a live vaccine in 1994 which has played a global role in AMPV control, thereby safeguarding the supply of poultry meat and eggs. Recent research and development at the UoL has identified key control measures, relating to vaccine application, vaccine selection, efficacy and safety, which have had a significant impact on poultry health and consequently, poultry producers and consumers. In particular, demonstration that live AMPV vaccines can revert to virulence, that vaccine type applied influences field protection and that continuous use of a single vaccine can influence circulating field strains, has resulted in UoL leading policy making with regard to current AMPV vaccine protocols.

Developing technologies for the control of viral diseases in cats

Summary of the impact

Viral infections pose a significant risk of long-term disease and death to cats. In Europe alone, over 30 million domestic cats are vaccinated each year against three core pathogenic viruses. Research performed at the University of Glasgow has systematically supported the development of key technologies against major feline viral diseases. This work has delivered incremental but wide-reaching benefits to veterinary healthcare and animal welfare by providing: (i) reagents used in the diagnostic industry; (ii) viral screening services for big cat conservation programmes; (iii) developmental input into the creation of one of the most efficacious and widely used vaccines against feline leukaemia virus; (iv) testing of feline vaccines for efficacy and safety; and (v) development of best practice guidelines and training for veterinary practitioners on feline viruses.

Control of Japanese Encephalitis

Summary of the impact

Globally the most important cause of encephalitis (inflammation and swelling of the brain) is the mosquito-borne Japanese encephalitis virus (JEV), which causes an estimated 70,000 cases annually across Asia. Although vaccines were developed years ago, their uptake in Asian countries has been hampered through lack of disease burden data, a consequence of poor surveillance, complicated diagnostics, and insufficient knowledge about disease outcomes. Research at the University of Liverpool has addressed each of these areas in turn, to overcome the roadblocks in vaccine implementation. The University of Liverpool (UoL), through its leading role on all the relevant WHO committees groups and meetings, has ensured that its research findings are translated through to impact by supporting new vaccination programmes across Asia. By 2013 vaccination had begun in 11 new countries, and the vaccine had reached more than 200 million people. The public health benefits, estimated from a health economic modelling study, are 854,000 cases and 214,000 deaths avoided, with an associated saving across Asia of US$ 1.024 billion.

Control Strategies towards eradication of Bovine Viral Diarrhoea

Summary of the impact

Interdisciplinary research at the Royal Veterinary College (RVC) has provided core evidence on which global efforts are based in order to eradicate one of the most economically damaging diseases of the cattle industry. The research findings have helped steer national programmes to eradicate Bovine Viral Diarrhoea (BVD) across Europe, South Asia and Australasia, reducing economic losses. Professor Joe Brownlie has additionally led pilot programmes in the UK, providing data for a national scheme, campaigned widely to highlight the issue and secured farming industry awareness and support through media exposure.

Improving feline health through the worldwide application of infectious and genetic disease polymerase chain reaction assays

Summary of the impact

Bristol University's School of Veterinary Sciences, a global leader in feline medicine, was the first UK centre to develop and commercially offer polymerase chain reaction (PCR) and quantitative (q) PCR assays to detect a range of feline infectious and genetic diseases. Since 2008 there has been a dramatic increase in the number of qPCR tests performed, with over 35,000 tests carried out between 2008 and 2013. The results of genetic testing have informed breeding programmes and resulted in a reduced prevalence of genetic disorders such as polycystic kidney disease (PKD). The results of testing for infectious diseases have informed diagnosis and treatment modalities and, together with the genetic testing, have contributed to significant improvements in feline health and welfare. This work has also generated commercial income in excess of £1.7M, which has been used to further research into feline infectious and genetic diseases.

Debunking MMR vaccine associated scares

Summary of the impact

Professor Rima's research on measles and mumps viruses over 4 decades at Queen's University allowed him to play an important role in re-establishing public confidence in the safety of the measles-mumps-rubella (MMR) vaccine. Claims that MMR vaccine could cause autism in 1998 undermined the vaccine uptake but Rima's expert testimony and that of others established in court that these claims were unfounded. This re-assurance and subsequent promotion of MMR vaccination reduced measles cases in the UK. In the USA, it also reduced the real risk that the Vaccine Court Fund, which compensates vaccinees for genuine vaccine related adverse events, would be bankrupted by over 50,000 claims amounting to between $30-50 Billion.

Translating research into novel immunotherapies delivers scientific and economic gains for the pharmaceutical/biotechnology sector in drug discovery

Summary of the impact

Research into novel immunotherapies has given rise to a novel drug (EtxB), which is now in Phase II clinical trials, and to a profitable contract research company partnering with the pharmaceutical industry to develop their compounds. Trident Pharmaceuticals was formed around patents filed by the University of Bristol, has received investment of [text removed for publication], successfully completed Phase I trials (2011) and is in the midst of Phase IIa trials in humans with inflammatory disease (2013). KWS BioTest arose as a result of the underpinning research and experience gained from developing EtxB, and is now a leading contract research organisation working with pharmaceutical and biotechnology companies developing novel treatments for human disease. KWS has directly contributed to the development of therapies at more than 75 different companies, employs 28 people, has exported [text removed for publication] and was 2012 winner of a Biomedical iNet Award for outstanding business achievement.

Improving fish health and the aquatic environment through vaccination

Summary of the impact

Research by the Institute of Aquaculture has made a significant contribution to the development of effective fish vaccines, some of which have been commercialised and are used widely within the aquaculture industry. The majority of farmed fish in the UK are vaccinated (44 million salmon and 7.5 million rainbow trout in 2012 alone) with vaccines developed at Stirling, resulting in vast improvements in survival and fish health, and a sustained minimal use in antibiotics through mass vaccination. Vaccines have been developed for all the major farmed species in Europe, and recently the first vaccine for Pangasius catfish in Vietnam (>2 million tonne).

"Shark-antibodies" as a platform for biologics drug development

Summary of the impact

Together the University of Aberdeen and Aberdeen city have become a major centre for biologics, the synthesis of medicines from compounds derived from living organisms. Commerce and industry have invested heavily in the process, creating specialist knowledge, jobs and an internationally-recognised network of expertise that promises further growth. This has arisen from ground-breaking research in Aberdeen into the VNAR antibody class that are the smallest binding sites so far identified in the animal kingdom and led to the validation of a new drug discovery platform. Spin-out companies were created (Haptogen Ltd, Cyclogenix Ltd and the pre-commercialisation vehicle Elasmogen) to exploit the emerging technology, which has completed successful efficacy trials in several animal models including late stage pre-clinical models, with trials in humans expected.

The claimed impact is therefore that: spin-outs have been created, highly skilled people have taken up specialist roles in companies; industry and venture money has been invested in patent protected research and development, business has adopted a new technology, award winning industry collaborations have been forged and jobs have been created.
